A physiological and biochemical approach to selecting the ideal diet for Ostrea edulis (L.) broodstock conditioning (part A) ArchiMer

To select the best microalgae for Ostrea edulis conditioning, four single species diets were fed to batches of broodstock, which were then compared using physiological and biochemical approaches. Ingestion and absorption were first studied, showing that Chaetoceros gracilis and Skeletonema marinoï were efficiently ingested (4.9–5.3 mg g−1 h−1) and absorbed (1.9–2.5 mg g−1 h−1) while Tetraselmis suecica led to the lowest physiological responses (0.36 and 0.12 mg g−1 h−1 respectively). When O. edulis were fed any microalgae except T-ISO with only 79 mg g−1, gonads accumulated carbohydrates from 116 to 141 mg g−1 and the extent of accumulation depended on the species supplied in the order T. suecica<C. gracilis<S. marinoï. Controlled reproduction of the European flat oyster requires the development of tools adapted to this species, including a practical anaesthesia and gonad sampling protocol to facilitate sex determination and the verification of gametogenesis. Three replicate groups of 10 oysters (mean weight +/- SD: 29.9 +/- 8.5 g) were anaesthetised in 5 L containers using magnesium chloride designed either for laboratory (Flucka (R)) or agricultural (Dead Sea Work (R): DSW) use, at two concentrations (50 or 72 g L-1). Depuis 1992, la surveillance de la santé des mollusques marins du littoral français est assurée par le réseau de Pathologie des Mollusques (Repamo). The present study describes some biological characteristics of sperm of the European flat oyster, a native species that has been over-exploited in the past and is nowadays classified as an endangered species in Europe.       The biology of gametes and larvae in brooding mollusc species remains little explored. The aim of this study is to present the results obtained in 2016, on biological characteristics of gametes and larvae of European flat oyster.       Measurements were carried out on 50 oysters, collected by divers in the Rade de Brest (Finistère, France), during the spawning period of this species. Gonad pH is acidic (6.58±0.54). After the dissociation of spermatozeugmata, an acellular structure to which the spermatozoa are clustered, the percentage of motile spermatozoa ranged from 20 to 34% and the Velocity of the Average Path from 70 to 85µm/s. The movement characteristics of Pacific oyster trochophores have received very little coverage in the scientific literature. Described here are not only changes in the swimming characteristics of Crassostrea gigas trochophores (size: 53-77 mu m) in relation to time after fertilization and to inter-female variation but also the effects of salinity and pH on movement characteristics. The percentage of motile trochophores was measured on images obtained through a dissecting microscope and the Velocity Average Path (VAP) was assessed using a Computer Assisted Sperm Analysis (CASA) system. The purpose of this study was to examine the responses of the oyster C. gigas to oxygen levels at subcellular and whole-organism levels. Two experiments were carried out. The first experiment was designed to measure at 15°C, 20°C and 25°C for 20 hours the clearance (CR) and oxygen consumption (OC) rates of oysters exposed at different concentrations of oxygen. Optimizing sperm cryopreservation in great scallop (Pecten maximus) ArchiMer

A basic cryopreservation protocol for great scallop sperm was recently published, with which thawed sperm survival remained limited to 25% of control values (Suquet et al. 2014). We designed here a set of five experiments to test ways of improving thawed sperm survival and storage by studying the effects of (1) cryoprotectant concentration; (2) sugar incorporation in the extender; (3) egg yolk addition to the extender; (4) sperm dilution rate; and (5) storage capacity of thawed sperm at 4 degrees C. Investigating the roles of chemical factors stimulating and inhibiting sperm motility is required to understand the mechanisms of spermatozoa movement. In this study, we described the composition of the seminal fluid (osmotic pressure, pH, and ions) and investigated the roles of these factors and salinity in initiating spermatozoa movement in the Pacific oyster. The acidic pH of the gonad (5.82 ± 0.22) maintained sperm in the quiescent stage and initiation of flagellar movement was triggered by a sudden increase of spermatozoa external pH (pHe) when released in seawater (SW). Depuis 2008, des mortalités massives d’huîtres creuses âgées de moins d’un an sont relevées sur le littoral français dès que la température de l’eau de mer atteint le seuil de 16°C. Ces mortalités de naissains sont associées à la détection du virus OsHV-1 μVar. The quality of oyster spermatozoa has become an issue for modern aquaculture. However, reliable protocols used to assess gamete quality are lacking. The aim of this work is to define a standardised experimental fertilisation protocol for Pacific oyster oocytes. Six experiments have been carried out in this study. The optimal conditions for Pacific oyster experimental fertilisation have been defined: (1) oocytes can be conserved in seawater for at least 4 h before fertilisation. Standardized experimental protocols designed to study Pacific oyster (Crassostrea gigas) gamete quality have not been previously published. Gamete sampling variations and confounding effect of interacting factors result in large variations between replicates, decreasing the effects of the studied factors. This work aims at defining a standardized procedure for incubation of oyster embryos designed for experimental purposes. In a first phase, four experiments were developed to improve embryo sampling and handling. Survival, Growth and Reproduction of Cryopreserved Larvae from a Marine Invertebrate, the Pacific Oyster (Crassostrea gigas) ArchiMer

This study is the first demonstration of successful post-thawing development to reproduction stage of diploid cryopreserved larvae in an aquatic invertebrate. Survival, growth and reproductive performances were studied in juvenile and adult Pacific oysters grown from cryopreserved embryos. Cryopreservation was performed at three early stages: trochophore (13 +/- 2 hours post fertilization: hpf), early D-larvae (24 +/- 2 hpf) and late D-larvae (43 +/- 2 hpf). From the beginning (88 days) at the end of the ongrowing phase (195 days), no mortality was recorded and mean body weights did not differ between the thawed oysters and the control.
